Abstract     
Testudinidae This study was conducted to determine the effect of lymph dilution on health parameters (packed corpuscular volume (PCV), white blood cell count, total protein (TP), albumin, globulin, total calcium, aspartate aminotransferase (AST), creatine kinase, glutamate dehydrogenase (GLDH), uric acid, phosphorus and urea, potassium, and sodium), ionized calcium and 25-hydroxycholecalciferol of tortoises (Testudo horsefieldi, T. gracea, T. marginata and T. hermanni; n=88). Blood (2 ml) samples were collected on the jugular vein and dorsal coccygeal vein of each animal immediately after hibernation and in the month of August. Lymph dilution was confirmed in the post-hibernation dorsal coccygeal vein samples. There were significant decreases in total calcium, albumin, 25-hydroxycholecalciferol, TP, and globulin in both sexes. Males had significantly lower urea, AST, CK and phosphorus while, the females had lower ionized calcium and uric acid levels. Blood values collected in August from the dorsal coccygeal vein were significantly lower than those from the jugular vein. The ionized calcium and total calcium of females were significantly higher than the males after hibernation and in August. On the other hand, the ionized calcium, total calcium, albumin and 25-hydroxycholecalciferol values in August were significantly higher than values after hibernation in males. It was also observed that 25-hydroxycholecalciferol were markedly reduced in dorsal coccygeal vein samples. It is concluded that jugular vein should be approached first before any sites when obtaining blood from Testudo sp. for diagnostic procedures. Extreme caution should be used in the subsequent interpretation of normality from lymph diluted samples.
