Abstract |
Ten polymorphic microsatellites were isolated from the red-eared slider, Trachemys scripta elegans, with a partial genomic library enriched for tandem repeat sequences of (CA)12 and (GA)12. For these markers polymorphism was investigated in 30 individuals collected from Yuehe pet market in Guangzhou, China. The allele number ranged from 5 to 12 per locus, with an average of 8.1. The observed and expected heterozygosities varied respectively from 0.4333– 0.9667 with an average of 0.7209, and 0.4452–0.8786 with an average of 0.7501, respectively. Exact tests for Hardy– Weinberg equilibrium (HWE) revealed that two of 10 loci significantly deviated from HWE after Bonferroni correction. One pair of loci (TSE14 and TSE79) showed significant linkage disequilibrium. The microsatellites developed here will be useful in population genetics and ecology in T. s. elegans.
